Description:

Glucosinolates는 십자화과 식물에 널리 존재하는 2차 대사산물의 일종입니다. 식물체 내에서는 안정적인 형태로 존재하지만, 식물이 물리적 손상을 입거나 생물학적 공격을 받으면 Myrosinase와 같은 효소에 의해 가수분해되어 이소티오시아네이트(Isothiocyanates), 황산염(Sulfates) 등 다양한 분해산물을 생성합니다. 이러한 분해산물은 항균, 병해충 저항성, 항산화 및 항암 활성 등 다양한 생물학적 특성을 나타냅니다.

Glucosinolates 함량을 정량적으로 측정하기 위한 분석 원리를 적용합니다. 미로시나아제가 글루코시놀레이트를 가수분해하여 포도당을 생성하고, 포도당은 Hexokinase(HK)와 Glucose-6-phosphate dehydrogenase(G6PDH)에 의해 순차적으로 대사됩니다. 이 과정에서 NADP⁺가 NADPH로 환원되며, NADPH는 1-mPMS를 통해 수소를 전달하여 WST-1을 formazan으로 환원시킵니다. 생성된 formazan은 450 nm에서 특징적인 흡광 피크를 나타내며, 이를 측정하여 시료 내 글루코시놀레이트 함량을 정량 분석할 수 있습니다.
